Chloride is an important intracellular ion, and changes in its concentration are associated with many physiological and pathological processes. Therefore, it is of great significance for the detection of chloride ions.

Optical probe is a tool that uses optical methods to detect biological samples, which has the advantages of non-invasiveness, high sensitivity, and high selectivity. In terms of chloride detection, the commonly used optical probes mainly include fluorescent dye-based optical probes and ratiometric fluorescent probes.

Among them, optical probes based on fluorochromes are made of certain fluorescent dyes that are chloride selective. When these dyes bind to chloride ions, their fluorescence properties change, enabling the detection of chloride ions. For example, a fluorescent dye based on rhodamine B, when it binds to chloride ions, its fluorescence intensity is weakened, so the concentration of chloride ions can be determined by detecting changes in fluorescence intensity.

In addition, optical probes based on ratiometric fluorescent probes are also commonly used chloride detection probes. Such a probe typically consists of two different fluorescent dyes, one of which emits fluorescence at the same wavelength as the other dye absorbs. When the probe binds to chloride ions, the fluorescence intensity of one of the dyes changes, resulting in a change in the ratio, enabling the detection of chloride ions. For example, a ratiometric fluorescent probe based on fluoride and cyanonine dyes, when it binds to chloride ions, the fluoride fluorescence intensity decreases while the fluorescence intensity of cyanine dyes increases, so the concentration of chloride ions can be determined by detecting the ratio of the fluorescence intensities of the two dyes.
