Principles of analysis.
Mycoplasma is cleaved and the released mycoplasma enzyme reacts with the substrate of the mycoplasma detection test kit to catalyze the conversion of ADP into ATP. Measure the pre- (Read A) and post-(Read B) ATP readings to determine whether mycoplasma contamination is present by the ratio of the before and after ATP readings. If there is no contamination, the reading after substrate addition is not much different from the reading before substrate is added, but in the case of mycoplasma contamination, the reading after substrate addition will differ by several times from the reading before substrate application (Figure 1). The new generation of MycoAlert Plus MycoAlert Plus produces a stronger light signal, so it can be used in plate and multi-function chemiluminescence instruments (Figure 2).
