Ferroptosis is an iron-dependent form of cell death, which is mainly characterized by the increase of ferrous and lipid peroxidation content in cells, which has been shown to play an important role in various biological processes such as development, aging, immunity, and cancer. Intracellular Fe3+ is reduced to Fe2+, which is stored in an unstable iron pool (LIP), and due to the instability and high reactivity of Fe2+, it reacts with polyunsaturated fatty acids on the cell membrane through the Fenton reaction to produce a large amount of lipid ROS, resulting in cell death.
Ferroptosis is tightly controlled by multiple regulators of iron metabolism, such as iron uptake, storage, utilization, and efflux. Studies have shown that various iron chelators can inhibit ferroptosis in cells, and conversely, iron supplementation accelerates ferroptosis. During ferroptosis, the ferrous content of cells tends to increase, so the detection of iron content in cells is also one of the commonly used detection methods for ferroptosis.
