Aldehyde PEG is an amine-reactive PEG derivative that can be used to modify biomolecules by available amine groups. The reaction between the aldehyde group and the -amine of the lysine residue and the -amine of the n-terminal produces an intermediate Schiff base. Further reduction with hydride will form a stable CN bond.
PEG aldehyde at 55 to 9Reacts with amine groups at pH 5. Higher pH will result in multiple pegylation of the terminal and lysine groups. Aldehyde PEG is a commonly used PEG reagent for N-terminal PEGylation in proteins or peptides. Reaction step: The target substance is dissolved in a polyethylene glycolation buffer.
Estimate the concentration of primary amine groups on the target material. Add the PEG aldehyde stock solution to the target conjugated material, and typically, 10 to 50 times more than the amount of the amine-containing material, the excess results in adequate conjugation, and the final concentration remains at least 10 mg ml. The mixture can be reduced by sodium borohydride by stirring for 2 4 h at room temperature, or overnight at 4 °C. Conjugates can be purified by size exclusion chromatography or dialysis.
