Introduction and synthesis method of PCl(5K)-PEG(1K)-Thiool.
PCL(5K)-PEG(1K)-ThioL is a biocompatible polymer composed of polycaprolactone (PCL), polyethylene glycol (PEG) and thiol groups. The uniqueness of this polymer lies in its amphiphilia, making it promising for a wide range of applications in drug delivery, tissue engineering, and biomedical applications.
Synthesis method. The synthesis of PCL(5K)-PEG(1K)-THIOL is mainly divided into three steps: the synthesis of PCL-PEG copolymer, the PEG-THIOL reaction and the deprotection reaction.
1.Synthesis of PCL-PEG copolymers: PCL and PEG are linked together by esterification. DCC or EDC are usually used as coupling agents to cause the carboxyl groups of PCL and PEG to react with amino groups to form amide bonds.
2.Peg-thiol reaction: In this step, the hydroxyl group of PEG is converted into a thiol group by reaction with a mercaptan. Commonly used thiols are maleimide, cysteine, etc. This step is usually performed under physiological conditions at pH to mimic the environment in an organism.
3.Deprotection reaction: In order to improve the water solubility of the polymer, the protective groups (e.g., BOC) on the PEG need to be removed. This is usually achieved by hydrolysis of weak acids or bases.
Applications & Prospects.
PCL(5K)-PEG(1K)-Thiool is widely used in drug delivery, tissue engineering, and biomedical applications due to its good biocompatibility and amphiphilicity. For example, it can be used as a drug carrier for targeted drug delivery; as a cell scaffold material for tissue engineering; and as a biosensor and diagnostic reagent, among others.
