With the continuous development of science and technology and the improvement of the suspect's awareness of anti-investigation, the biological samples left at the scene have been decreasing, showing a trend of miniaturization, and it is almost impossible to conduct secondary experiments. However, the storage and transportation conditions of conventional forensic DNA detection reagents are harsh, and if they are not stored properly, the sensitivity of the reagents is easy to decrease or even fail, resulting in the waste of samples. In this case, the traditional detection reagent can no longer meet the needs of the investigation, so the existing detection reagent needs to be improved. As one of the latest solutions, lyophilized reagents have been widely recognized and applied.

In the experimental validation part, the article lists a number of experimental data and cases, which fully demonstrate the accuracy and reliability of lyophilized reagents in practical applications. These data are derived from extraction and amplification experiments on different types of micro-degraded samples, including highly degraded and complex samples containing inhibitors. The experimental results show that the reagent can obtain complete STR typing results in these samples with good reproducibility.
In the future, lyophilized reagents are expected to become one of the mainstream tools in the field of forensic technology, providing strong technical support for solving cases and judicial fairness.
In this paper, it is pointed out that for micro samples, the volume of lyophilized reagent should be increased(10 15 l lyophilized in situ).It is one of the ways to improve the detection rate.
