Bronchial asthma (asthma) is a chronic inflammatory disease of the airways that seriously endangers human health. The latest epidemiological survey in China shows that the prevalence of asthma in people aged 20 years and above is 42%。"Severe asthma accounts for less than 10% of them, but it is very problematic, with airway remodeling being the main cause and its pathogenesis still unelucidated. According to Lai Tianwen, breakthroughs are urgently needed in research on the pathogenesis of asthma, new biomarkers and potential intervention targets.
In recent years, the pathogenesis of asthma induced by environmental exposures such as allergens, microorganisms and air pollutants mediated by epigenetic modifications has attracted much attention. Apparent modifications mainly include acetylation, methylation, phosphorylation and ubiquitination, among which histone acetylation is particularly important. SIRT6, as a member of the sirtuin deacetylase family, plays a leading role in regulating cell proliferation, stress response, genome stability, energy balance, and aging.
Lai's team found that SIRT6 expression was positively correlated with airway remodeling and disease severity in asthma patients. SIRT6 of the airway epithelium** acts as an epigenetic regulator to exacerbate the airway inflammation response and airway remodeling in asthma by promoting IL-17A-mediated inflammatory chemokine and mesenchymal reprogramming.
